As of 2018, she has been diagnosed with Type 2 diabetes, making her one of the few diabetic chefs in the United States. The disease which generally affects people over 50 is linked to poor blood glucose control, and increased risk of heart disease and stroke. However, some people with the condition have no immediate symptoms. Deen says she is "not ashamed" to have the condition, and has said that she can kick her Type 2 diabetes by eating healthily and taking better care of herself. She has remarked that her diabetes is "a natural thing" and has referred to herself as a "diabetic". In April 2017, Deen took a voluntary two-week leave of absence from her cooking show to deal with the condition.

Deen has three children by her first marriage to Pittman; a daughter, Savannah, from a previous relationship, and twins Anthony and Adrienne; and a son, Blake. Deen and her third husband, musician and restaurateur Bob Wood, have a daughter, Zellyn, who was born in 2007. She also has step-children Brandon, Katie, and Ty, and has a grand daughter named Ava.
Deen is an evangelical Christian.
